| اختلالات هموستاتیک منجر به ایجاد لخته‌های نابجا و ترومبوآمبولی می‌گردد. داروهای فیبرینولیتیک نظیر استرپتوکیناز و اوروکیناز برای درمان ترومبوآمبولی بکار می‌روند. این داروها محدودیت‌هایی دارند و سبب ایجاد اثرهای ناخواسته و گاهی مرگبار می‌شوند. ترکیبهای گیاهی به‌عنوان ترکیبهایی سالم و بدون اثر جانبی در نظر گرفته می‌شوند. با نظر به اینکه چهار گیاه زردچوبه (Curcuma domestica Valet.)، آویشن شیرازی (Zataria multiflora Boiss.)، دارچین (Cinnamomum verum J.Presl) و گلپر (Heracleum persicum Desf. Ex Fischer) در طب سنتی به‌عنوان گیاهان ضد انعقاد و رقیق‌کننده خون معرفی شده‌اند، در این مطالعه اثرهای فیبرینولیتیک این گیاهان مورد بررسی قرار گرفت. جهت بررسی اثر فیبرینولیتیک، فیبرینوژن نشاندار شده با FITC با پلاسمای انسانی مخلوط و به آن ca+2 اضافه گردید. عصاره پلی‌فنلی تهیه شده چهار گیاه ذکر شده در غلظت‌های 05/0، 5/0، 5 و 50 (mg/ml) به تنهایی و همراه با استرپتوکیناز به لخته نشاندار اضافه شدند و 10، 20، 40 و 60 دقیقه پس از اضافه نمودن عصاره‌ها فلورسانس ناشی از FITC تعیین گردید (510Em= و 478Ex=). نتایج نشان می‌دهند که عصاره‌‌‌‌‌‌‌‌‌‌‌‌‌های آویشن شیرازی، دارچین و زردچوبه دارای اثر فیبرینولیتیک می‌باشند. این اثرها معنی‌دار (05/0p≤) و وابسته به دوز می‌باشند. عصاره گلپر اثر فیبرینولیتیک معنی‌داری ندارد. اثر فیبرینولیتیکی ناشی از استرپتوکیناز در حضور عصاره آویشن شیرازی به صورت وابسته به دوز افزایش می‌یابد. در حضور عصاره زردچوبه و دارچین فقط در غلظت‌های 5 و 50 (mg/ml) اثر فیبرینولیتیکی ناشی از استرپتوکیناز به صورت معنی‌دار افزایش می‌یابد (05/0p≤). عصاره گلپر تأثیری بر اثر فیبرینولیتیک استرپتوکیناز ندارد. بنابراین می‌توان گفت که آویشن شیرازی دارای بیشترین اثر فیبرینولیتیک و پس از آن به‌ترتیب دارچین و زردچوبه دارای اثر فیبرینولیتیک معنی‌دار هستند و می‌توانند در شرایط آزمایشگاهی سبب حل لخته شوند. در پایان، پیشنهاد می‌شود که ترکیبهای موجود در این گیاهان که سبب حل لخته می‌شوند بررسی و همچنین خصوصیت حل لخته توسط این گیاهان در محیط In vivo در تحقیقات آینده شناسایی شوند. | ||

| Disturbance in haemostatic system causes abnormal clots in vessels and thromboambolia. Streptokinas and urokinas are used for emergency treatment of thromboambolia. These drugs have certain limitations which cause serious and sometimes fatal consequences. Herbal preparations are considered to be safe and without side-effects. Since in ancient times it was reported that Zataria moltiflora Boiss., Curcuma domestica Valet., Cinnamomum verum J.Presl and Heracleum persicum Desf. Ex Fischer present anticoagulant effects. This study performed to evaluate fibrinolytic effects of these plants. To evaluate fibrinolytic effects, labeled fibrinogen with FITC and Ca2+ was added to human plasma. Polyphenolic extracts (0.05, 0.5, 5, 50mg/ml) without and with streptokinas, were added to labeled clot. Fluorescence was determined after 10, 20, 40 and 60 minutes (Ex=478, Em= 510). Our findings show that extract of Zataria moltiflora, Curcuma domestica and Cinnamomum verum reveal fibrinolytic effects (P<0.05). This effects are significant and dose dependable, but Heracleum persicum had no significant fibrinolytic effect (P>0.05). Fibrinolytic effects of streptokinase in presence Zataria moltiflora are increased significantly and dose dependably. This effect in presence of Curcuma domestica and Cinnamon only in 5 and 50 mg/dl are increased. Heracleum persicum does not affect fibrinolytic properties of streptokinase. The present study indicate that, Zataria moltiflora presents best fibrinolytic effect, then Curcuma domestica and Cinnamon verum respectively reveal significant fibrinolytic effect and could lysis blood clots in vitro. Consequently, we suggest that further studies to determine in vivo clot dissolving properties and active component(s) of these herbs for clot lysis are warranted. | ||
