| چکیده ﺳﺎﺑﻘﻪ و ﻫﺪف: آنیسون یا بادیان رومی با نام علمی Pimpinella anisum L.از تیره (Apiaceae (Umbelliferea و جنس Pimpinella است. آنیسون بومی نواحی سواحل غربی دریای مدیترانه، مصر و آسیای صغیر است. جنس Pimpinella دارای گونههای متعددی است که تاکنون ۱۵۰ گونه در اروپا، آسیا و آفریقا متعلق به این جنس شناخته شدهاند. حدود 2 تا 3 درصد وزن بذر آنیسون را اسانس تشکیل می‌دهد و مهمترین ماده تشکیل دهنده اسانس، ترانس آنتول به میزان 80 تا90 درصد آن است. این مطالعه با هدف بهینهسازی باززایی این گیاه در محیط درون شیشهای انجام شده است. ﻣﻮاد و روشﻫﺎ: در آزمایش اول، جوانهزنی بذرهای گیاه و انتخاب روش مناسب ضدعفونی برای آلودگی‌زدایی نمونهها بررسی شد. در آزمایش دوم، باززایی مستقیم گیاه تحت تأثیر ترکیبات مختلف BA، Kin و NAA روی ریزنمونه جوانه هیپوکوتیلی بررسی گردید. در آزمایش سوم، تولید کالوس از ریزنمونههای ریشه، هیپوکوتیل، جوانه هیپوکوتیلی، برگ لپهای و برگ معمولی با استفاده از غلظتهای مختلف 2,4‐Dانجام شد. در آزمایش چهارم، اثر دامنه محدودتری از غلطتهای مناسب از 2,4‐D روی ریزنمونههای دمبرگ و ریشه بررسی شد. در آزمایش پنجم، کالوسهای تولید شده در مرحله قبل برای باززایی در معرض غلظتهای مختلف BAو NAA قرار گرفتند. همه این آزمایش‌ها در قالب طرح‌های کاملاً تصادفی با سه تکرار انجام شد. آنالیز دادهها نیز با استفاده از نرمافزار Minitab انجام و مقایسه میانگین تیمارها با آزمون توکی در سطح احتمال 5 درصد انجام شد. یافته‌ها: بهترین پاسخ جوانهزنی به میزان 70 درصد در شرایط 24 ساعت خیس کردن بذرها و ضدعفونی با الکل 70 درصد و هیپوکلریتسدیم 5/1 درصد در محیط کشت MS 2/1 بدست آمد. بهترین پاسخ باززایی مستقیم از نظر طول دمبرگ مربوط به تیمارهای 9 (5/0 میلیگرم بر لیتر BA به اضافه 5/0 میلیگرم بر لیتر Kin به اضافه 5/0 میلیگرم بر لیتر NAA )، 14( 5/0 میلیگرم بر لیتر Kinبه اضافه یک میلیگرم بر لیتر NAA)، 17( 25/0 میلیگرم بر لیتر BA به اضافه 25/0 میلیگرم بر لیتر Kin به اضافه یک میلیگرم بر لیتر NAA ) و 19 (یک میلیگرم بر لیتر BA به اضافه یک میلیگرم بر لیتر Kin به اضافه یک میلیگرم بر لیتر NAA ) با حدود 4 سانتیمتر بود. از نظر تعداد برگ، بهترین پاسخ مربوط به تیمارهای 14و 17 با 47 عدد برگ بود. در آزمایش سوم، ریزنمونههای دمبرگ و ریشه بهترین کالوسزایی را داشتند. در آزمایش چهارم، با استفاده از تیمار 5/0 میلیگرم بر لیتر BA به اضافه 75/0 میلیگرم بر لیتر 2,4D به میزان 22/97 درصد در ریزنمونه دمبرگ، کالوسزایی مشاهده شد. در آزمایش پنجم، کالوسهای بدستآمده از مرحله قبل در تیمار شاهد بدون هورمون بیشترین شاخهزایی را به میزان 40 درصد نشان دادند. بعضی از شاخه‌ها ریشه‌دار شده و گیاهچههای حاصل پس از سازگاری به گلخانه منتقل شدند. ﻧﺘﯿﺠﻪگیری: در این آزمایش اگرچه باززایی مستقیم منجر به تولید ساقه نشد، اما تولید برگ در این روش مشاهده شد. با توجه به عدم مشاهده نتایج مورد انتظار در روش باززایی مستقیم، تولید کالوس و باززایی غیرمستقیم مدنظر قرار گرفت که با موفقیت چشمگیر همراه بود. در مجموع نتیجه‌گیری شد که برای باززایی گیاه انیسون، روش غیرمستقیم یعنی باززایی گیاه از کالوس مناسبتر از روش باززایی مستقیم است. دمبرگ به عنوان بهترین ریزنمونه معرفی شد. تیمار هورمونی 75/0 میلیگرم بر لیتر 2,4-D به همراه 5/0 میلیگرم بر لیتر BA توانست در ریزنمونه دمبرگ به میزان 22/97 درصد کالوس القا نماید، بنابراین به عنوان یک ترکیب هورمونی موفق برای کالوسزایی در انیسون معرفی شد. تولید شاخه و ریشه از کالوس در تیمار شاهد، نشان داد که بعد از بدست آوردن کالوس، بدون استفاده از تنظیم‌کننده‌های رشد گیاهی میتوان به گیاه کامل دست پیدا کرد. نتایج این پژوهش میتواند در مطالعات ریزازدیادی، مهندسی ژنتیک، کشت سوسپانسیون سلولی و تولید متابولیتهای ثانویه و سایر پروژههای مرتبط با اصلاح گیاه و پروژههای کشاورزی مولکولی مورد استفاده قرار گیرد. با توجه به ارزش دارویی گیاه آنیسون، نتایج این پژوهش میتواند دارای کاربرد عملی باشد. | ||

| Extended Abstract Background and objectives: Anise Pimpinella anisum L. is a valuable medicinal plant that 2-3% of its seed weight is essential oils, and about 80-90% of its essential oil is trans-antol. This research was carried out to optimize the indirect organogenesis of P. anisum under in vitro conditions. Methodology: In the first experiment, different methods of seed germinating and disinfection treatments for decontamination of samples were investigated. In the second experiment, the effect of different combinations of BA, Kin and NAA on the direct regeneration of hypocotyl bud explants was investigated. In the third experiment, callus production was investigated from explants of root, hypocotyl, hypocotyl bud, cotyledon leaf and normal leaf, using different concentrations of 2,4-D. In the fourth experiment, the effect of a narrower range of 2,4-D concentration on the callus formation of petiole and root explants was evaluated. In the fifth experiment, the produced calli was exposed to different concentrations of BA and NAA for regeneration. All experiments were conducted in a completely randomized design with three replications. Data analysis was done using Minitab software, and the mean comparison was made using the Tukey method (p<0.05). Results: The best germination was obtained at a value of 70% in 24 hours of seed soaking in water (hydro priming) and disinfection with 70% alcohol and 1.5% sodium hypochlorite in 1/2 MS culture medium. In direct regeneration, stem-less leaves were produced. The best direct regeneration response in terms of petiole length was related to treatments of 9 (0.5 mg/L BA plus 0.5 mg/L Kin plus 0.5 mg/L NAA), 14 (0.5 mg per liter Kin plus 1mg/L NAA), 17 (0.25 mg/L BA plus 0.25 mg/L Kin plus 1mg/L NAA) and 19 (1mg/L BA plus 1mg/L Kin plus 1mg/L NAA) with about 4 cm. The highest leaf number (47 leaves) was obtained using the treatments of 14 and 17. In the third experiment, petiole and root explants had the best callus formation. In the fourth experiment, using the treatment of (0.5 mg/L BA +0.75 mg/L of 2, 4-D), the highest rate of 97.22% callus formation was observed in petiole explants. In the fifth experiment, the callus obtained from the previous stage induced the highest shoot with a value of 40% in the control treatment without hormones. Some branches were rooted, and the resulting seedlings were transferred to the greenhouse after adaptation. Conclusion: It was concluded that for the regeneration and breeding of anise plant, the indirect regeneration of the callus is more suitable than the direct regeneration method. Also, the petiole was a suitable organ as the initial explant. The 1.5% sodium hypochlorite was the best disinfectant for seed decontamination. Hormonal treatment of 0.75 mg/L of 2, 4-D +0.5 mg/L of BA induced the highest rate (97.22%) of callus in a petiole explant. Therefore, it could be used as a successful hormonal compound for callus formation in anise. The production of branches and roots from callus in the control treatment indicated that it was possible to obtain a whole plant without using any plant growth regulators. The results of this research can be used in micropropagation studies, genetic engineering, cell suspension culture and production of secondary metabolites and other projects related to anise improvement and molecular studies. | ||
