| The genus Seidlitzia has four herbaceous species viz. S. florida (M.B.) Bge. ex Boiss., S. cinerea (Moq.) Bge. ex Botsch. and S. stokii that are herbaceous, and S. rosmarinus (Ehrenb.) Bge. ex Boiss. is shrubby belong to the family Amaranthaceae. In the present study, ISSR molecular markers were used to evaluate the genetic diversity among 85 plant samples from 17 populations of S. rosmarinus. Four random primers were used to perform ISSR reactions and in total 221 DNA bands were obtained. After forming 0 and 1 matrix with the help of GenAlex and NTSYS software, it was used to interpret ISSR analysis data. Molecular analysis of variance (AMOVA) was performed to estimate genetic diversity out of 85 individuals. Of total diversity, the highest amount of genetic diversity (68%) was related to intra-population diversity, 28% of inter-population diversity, and 4% of inter areas was calculated. MCMC analysis using Structure 2.3.4 software was used to investigate the genetic diversity populations of S. rosmarinus. Main coordinates analysis by the PCOA method was also used for extract maximum information from molecular data. | ||

| جنس Seidlitzia دارای چهار گونه در ایران است. گونه S. rosmarinus به شکل درختچهای و سه گونه دیگر علفی هستند. هدف از این تحقیق، بررسی تنوع ژنتیکی و ساختار جمعیتی 85 نمونه گیاهی 17 جمعیت گونه S. rosmarinus و پی بردن به شناسایی احتمالی فرمهای تاکسونومیکی پایینتر از سطح گونه با استفاده از نشانگر مولکولیISSR بود. از 16 پرایمر عمومی و چهار پرایمر اختصاصی استفاده شد که در مجموع 221 باند ایجاد کردند. دندروگرام دادههایISSR مربوط به 17 جمعیت براساس ماتریس تشابه به روشUPGMA و ضریب تشابهDice از 40/0 تا 96/0 متغیر بود. براساس آنالیز واریانس مولکولی (AMOVA)، بیشترین تنوع (68%) تنوع درون جمعیتی، 28% تنوع بین جمعیتی و 4% مربوط به تنوع بین مناطق بود. آنالیزMCMC به منظور بررسی ساختار ژنتیکی در جمعیتهای گونهS. rosmarinus مورد بررسی قرار گرفت. همچنین، آنالیز مختصات اصلی به روشPCOA براساس محور مختصات اصلی اول و دوم برای نشانگرISSR انجام شد که این روش به عنوان روش مکمل برای تجزیه خوشهای با هدف استفاده بهینه و استخراج حداکثری اطلاعات از دادههای مولکولی ارایه شد. نتایج به دست آمده از نشانگرISSR از درجه اطمینان بالایی برخوردار بود. نتایج به دست آمده با استفاده از نشانگرISSR نشان داد که نسبت به سایر نشانگرها دقت بیشتری داشته و روابط بین جمعیتها را با وضوح بهتری نمایان ساخت. از طرفی، فرمهای تاکسونومیکی احتمالی پایینتر از سطح گونه با توجه به اختلافات کم بین جمعیتها مشاهده نشد. | ||
