Effect of Sodium Cyanide-induced Tissue Hypoxia on Reproductive Capability of Male Mice and the Protective Effect of Ethyl Pyruvate | ||

| The present study aimed to assess the effect of tissue hypoxia induced by sodium cyanide (NaCN) on male mice fertility and the protective role of ethyl pyruvate (EP). A number of 30 adult mice were assigned to three groups: 1) a control group, 2) a treatment group treated with 2 mg/kg of NaCN, and 3) a treatment group treated with 2 mg/kg of NaCN, along with 40 mg/kg EP (NaCN+EP). After 35 days, animals were anesthetized and serum, sperm, and tissue samples were taken. The results demonstrated a significant decrease in sperm quality, reproduction potency, and anti-oxidant potential, as well as an increase in lipid peroxidation in the NaCN group (p <0.05). Moreover, the use of EP effectively restrained the disastrous effects of tissue hypoxia. It can be concluded that EP can moderate the complications resulting from tissue-hypoxia that is related to testes parameters. | ||

| بررسی اثر هایپوکسی بافتی ناشی از سدیم سیانید بر توان باروری موش سوری نر و نقش محافظتی اتیل پیروات | ||

| کاهش میزان اکسیژن رسانی میتواند موجب اختلال در عملکرد بیضه و اسپرماتوژنز گردد. هدف این مطالعه بررسی اثر هیپوکسی بافتی ایجاد شده توسط سدیم سیانید بر توان باروری موش سوری نر و نقش محافظتی اتیل پیروات بر آسیب ناشی از این هیپوکسی بود. از این رو در تحقیق حاضر تعداد 30 سر موش سوری نر بالغ 25 تا 30 گرمی در 3 گروه مورد استفاده قرار گرفتند. گروه کنترل دریافت کننده سرم فیزیولوژی، گروه دریافت کننده سدیم سیانید با دوز mg/kg 2 هر 24 ساعت یک بار به صورت تزریق داخل صفاقی و گروه دریافت کننده سدیم سیانید با دوز mg/kg 2 به همراه اتیل پیروات با دوز mg/kg 40 به صورت تزریق روزانه داخل صفاقی به مدت 35 روز دریافت نمودند. پس از پایان دوره تجویز، پس از بیهوشی خونگیری از طریق قلب انجام و متعاقب آسان کشی نمونههای سرمی جهت آزمایشات سرمی، نمونههای اسپرم جهت آزمایشات بررسی اسپرم و IVF و نمونههای بافتی نیز پس از جدا سازی در داخل محلول بوئن فیکس شده و پس از انجام برشهای پارافینی با رنگ آمیزیهای هماتوکسیلین- ائوزین مورد بررسی هیستومورفومتری قرار گرفتند. نتایج نشان دهنده کاهش معنیدار کیفیت اسپرم و توان باروری، کاهش فاکتورهای TDI ، RI و SI و تعداد سلولهای لیدیگ، کاهش توان آنتی اکسیدانتی و افزایش میزان پراکسیداسیون چربیها درگروههای دریافت کننده سدیم سیانید بود (05/0>P). این در حالی بود که استفاده از اتیل پیروات به عنوان یک آنتی اکسیدان قوی به خوبی توانسته بود اثرات سوء حاصل از هیپوکسی بافتی را جبران کند. بنابراین میتوان نتیجه گرفت که اتیل پیروات قادر به خنثی نمودن عوارض ناشی از هیپوکسی بافتی در رابطه با پارامترهای بیضه میباشد. | ||
