| اکثریت ویروس‌های عضو جنس Vitivirus از گیاه مو (Vitis vinifera) جداسازی شده‌اند. از بین این ویروس‌ها، Grapevine virus A-GVA، Grapevine virus B-GVB و GVF Grapevine virus F- از ایران گزارش شده‌اند (1; 2; 3). در این تحقیق، حضور ویروس D انگور (Grapevine virus D-GVD) در تاکستان‌های استان آذربایجان غربی بررسی و گزارش گردید. در سال 1396 تعداد 33 نمونه از تاک‌های با علائم مشابه با آلودگی ویروسی شامل پیسک برگ، بدشکلی ساقه و نیز تغییر رنگ بافت ساقه از آذربایجان‌غربی جمع‌آوری و آر.ان.ای کل از بافت دمبرگ و رگبرگ‌های اصلی استخراج گردید. از آغازگرهای اختصاصی ناحیه ژن پروتئین پوششی GVD (4) برای ردیابی ویروس استفاده شد. پس از آزمون RT-PCR در سه نمونه یک قطعه DNA به طول حدود 470 جفت باز تکثیر گردید. محصول واکنش PCR در مورد دو نمونه (Irn-AzW7 و Irn-AzW23) خالص‌سازی و تعیین توالی شدند. توالی‌های بدست آمده تحت رس‌شمار MT133689 و MT152314 در بانک ژن جهانی (GenBank) ثبت شده و با استفاده از ابزار BLASTn با توالی‌های موجود در GenBank (قابل دسترس در وبگاه NCBI) مقایسه گردید. بر اساس نتایج حاصل توالی بدست آمده در مورد دو جدایه Irn-AzW7 و Irn-AzW23 دارای مشابهت 3/94 درصد با جدایه ایتالیایی GVD (رس شمار Y07764) و 0/92 درصد با جدایه ترکیه‌ای TR167 (رس شمار KY689027) بود. این ویروس تاکنون از انگور در کشورهای ایتالیا، تونس، ترکیه، کرواسی از قاره اروپا و برزیل در آمریکای جنوبی گزارش شده است (4; 5; 6). این اولین گزارش از وقوع آلودگی GVD در ایران می-باشد. | ||

| Most virus members of the Vitivirus genus have been isolated from grapevine (Vitis vinifera), among which Grapevine virus A (GVA), Grapevine virus B (GVB) and Grapevine virus E (GVE) have been reported from Iranian vineyards (1, 2, 3). In this study, the occurrence of Grapevine virus D (GVD) has been reported from Iran. During a survey in 2017, a total number of 33 symptomatic samples were collected from grapevines showing leaf mottling, stem deformations and color change of stem tissue in Azarbaijan-e-gharbi province. Total RNA was extracted from leaf petiole and primary veins and was tested for the presence of GVD by RT-PCR using primers specific to the coat protein gene of GVD (4). A DNA fragment with the expected size of 470 bp was amplified from three samples. The amplified DNA fragments of two samples (Irn-AzW7 and Irn-AzW23) were purified and their nucleotide sequence were determined. The nucleotide sequence obtained from PCR amplicons was submitted in the GenBank under accession numbers MT133689 and MT152314 and was subjected to BLASTn analysis (available at NCBI) to confirm the identity of the target virus. The Iranian isolates showed 94.3 % nucleotide sequence identity with a GVD isolate from Italy (Ac.No. Y07764) and 92 % with a GVD isolate from Turkey (Ac.No. KY689027). GVD has been reported from Italy, Tunisia, Turkey, Croatia in Europe, and Brazil in South America (4, 5, 6). To our knowledge, this is the first report on the occurrence of GVD in Iran. | ||
