بررسی اثر عصاره الکلی بذر گیاه تاتوره بر درد حاصل از آزمونهای فرمالین و صفحه داغ در موشهای صحرایی نر | ||

| گیاه تاتوره در کتابهای پزشکی کهن ایران و در سالهای اخیرا توسط محققان امروزی گونه‌های خاصی از این گیاه به عنوان گیاه ضد درد معرفی شده است. مشخص شده است که قسمتهای مختلف این گیاه بخصوص بذر آن حاوی آلکالوئیدهای مختلف می‌باشد. با توجه به تداخل سیستم اوپیوئیدی با مواد مختلف و از جمله مهمترین آنها آلکالوئیدها یا مواد قلیایی و نقش واضح سیستم اوپیایی در کنترل درد، تحقیق حاضر به مطالعه اثر عصاره الکلی بذر گیاه تاتوره به عنوان غنی‌ترین جزء این گیاه در تسکین دردهای حاد و مزمن پرداخته است. برای ایجاد دردهای حاد و مزمن یا التهابی از آزمونهای صفحه داغ و تزریق فرمالین استفاده شد. برای بررسی اثر بذر گیاه، پس از عصاره‌گیری از بذر گیاه، عصاره از طریق داخل صفاقی تزریق و بدنبال آن آزمونهای درد صورت می‌گرفت. با اعمال میزان های متفاوت عصاره در دو نوع آزمون فوق میزان های موثر (ED50 ) 50 و 25 میلی‌گرم به ازای هر کیلو گرم به ترتیب در آزمون فرمالین و صفحه داغ بدست آمد. میزان های موثر این گیاه در دو نوع آزمون در گروه تحت درمان با عصاره حدود 20 دقیقه قبل از آزمون درد تزریق و آزمون درد انجام می‌پذیرفت. میزان درد موشهای عصاره گرفته با میزان درد موشهای کنترل (بدون دریافت عصاره) با کمک آزمون Student t-test تحلیل و با 05/0P< معنی‌دار بودن تفاوت درد در دو گروه در نظر گرفته شد. نتیجه آزمون آماری داده‌های مطالعه ما نشان داد که میزان درد حاد و مزمن حاصل از آزمون فرمالین و همچنین درد مرحله یک حاصل از آزمون صفحه داغ به میزان قابل توجهی توسط عصاره الکلی گیاه تاتوره کاهش می‌یابد. علاوه بر این با اعمال میزان های افزایشی تاتوره از mg/kg4000- 500 به شکل داخل صفاقی میزان مرگ و میر موشها بررسی شد، که نتیجه آن میزان کشنده متوسط (LD50) حدود mg/kg 2500 بدست آمد. به طور کلی نتایج آزمایش های ما با عصاره الکلی گیاه تاتوره نشان داد که درد حاد و مزمن حاصل از آزمونهای فرمالین و صفحه داغ توسط عصاره الکلی این گیاه به شکل معنی‌داری کاهش پیدا می‌کند. همچنین با توجه به فاصله زیاد بین میزان موثر و میزان کشنده، شاید بتوان بذر گیاه تاتوره را به عنوان ماده ضد درد معرفی کرد. | ||

| The medicinal plant Datura stramonium (D.s) is a rich source of alkaloids which has been introduced as an analgesic herb. Regarding the possible interaction between opioid and muscarinic systems and the key role of opioid receptors in alleviation of pain, we investigated the effect of alcoholic Datura seed extract in acute and chronic pain. In the present study in order to producing acute and chronic pain, hot plate and formalin tests were used. In treatment group 20 min prior to pain scoring the extract was intraperitonealy administered to the animals and the results was compaired with the control group. In our experiments, the extract alleviated the pain dose dependently, and ED50 was equal to 25 and 50 mg/kg in hot plate and formalin tests, respectively. Analysis of the data by student t-test have shown that the acute and chronic pain producing by formalin and hot plate tests were significantly attenuated in animals which treated with DS seed extract. In order to determine the LD50, doses of 500-4000 mg/kg i.p. of the extract were applied to the animals. In this experiment the calculated LD50 was 2500 mg/kg. In addition, our results show that the DS seed extract has an analgesic effect in both acute and chronic pain which was produced by hot plate and formalin tests. It is likely that, this effect can be attributed to the D.s extract alkaloids which interacted with opioid system. Also, regarding the significant difference between ED50 and LD50, perhaps we can introduce the D.s seed extract as an analgesic medicinal plant. | ||
