| فلاونوئیدها ترکیبهایی پلی‌فنولیکی هستند که در همه غذاها با سرچشمه گیاهی وجود دارند. آثار آنتی‌اکسیدانی آنها به اثبات رسیده است، به‌طوری که مصرف سبزیجات و گیاهان از بروز سرطان و بیماری‌های قلبی و عروقی نظیر آترواسکلروز جلوگیری می‌کند. از طرفی امروزه مصرف آنتی اکسیدان‌های سنتتیک به دلیل سمیت آنها محدود گشته است و توجه جوامع پزشکی به استفاده و یافتن آنتی اکسیدان‌های طبیعی معطوف گشته است. اثرات آنتی اکسیدانی یک ترکیب توسط روشهای مختلف مورد مطالعه قرار می‌گیرد یکی از آنها بررسی اثرات یک ترکیب بر حفاظت گلبول قرمز از همولیز می‌باشد.در این مطالعه اثرات فلاونوئیدهای کامفرول،کوئرستین، مورین، روتین بر همولیز گلبول‌های قرمز و تأثیر آنها بر ظرفیت -SH غشاء گلبول‌های قرمز به عنوان شاخص محافظت از غشاء مورد بررسی قرار گرفته است. در ابتدا محلول‌های فلاونوئیدی تهیه گردید. میزان همولیز گلبول‌های قرمز و نیزظرفیت -SH غشاء گلبول‌ها با استفاده از روش اسپکتروفتومتری تعیین گردید جهت القاء پراکسیداسیون گلبول‌های قرمز از2 و'2 آزوبیس (2 آمیدینو پروپان) دی‌هیدروکلرید (AAPH) استفاده شد. اثرهر فلاونوئید بر همولیز گلبول‌های قرمز خون درسه غلظت (10 ، 5 و 0.5) میکروگرم بر میلی‌لیتر مورد بررسی قرار گرفت. در مورد تأثیر بر میزان گروه‌های -SH تنها بالاترین غلظت یعنی 10 میکروگرم بر میلی‌لیتر از هر فلاونوئید استفاده گردید. در تمامی موارد خاصیت آنتی‌اکسیدانی با افزایش غلظت، افزایش یافته است بیشترین تأثیر بر مهار همولیز گلبول قرمز را روتین به میزان 5/42 درصد داشته است همچنین کامفرول، روتین و مورین باعث حفاظت‌گروه‌های -SH به ترتیب به میزان 6 درصد و 233 درصد و 26.4 درصد گردیده‌اند. نتایج حاصل نشان می‌دهد که می‌توان از فلاونوئیدهای مزبور و گیاهان حاوی آن به عنوان مواد آنتی‌اکسیدان طبیعی در پیشگیری و یا درمان بسیاری از بیماری‌هایی‌که پاتوژنز آن پراکسیداسیون لیپید باشد استفاده نمود. | ||

| Flavonoids which are polyphenolic substances are found in different vegetable and fruits and they have anti-oxidant properties. Epidemiological studies showed that flavonoids reduce the prevalence of cardiovascular disease. On the other hand, the usage of synthetic antioxidants have been limited because of their toxicity and, there are different researches to find better natural antioxidants. This survey investigates the effect of some pure flavonoids such as kaempferol, quercetin, morin and rutin on red blood cell (RBC) hemolysis and their -SH capacity as membrane protection indicator. The flavonoids solutions were prepared. Rate of RBC hemolysis and -SH capacity of cell membrane were determined by spectrophotometer. 2,2´azobis2 amidino propane dihydrechloride (AAPH) was used to induce RBC peroxidation. The effect of each flavonoids on RBC hemolysis was examined in 3 concentrations (0.15, 5, 10) μg/ml but for investigating the effect on -SH groups only the highest concentration (10 μg/ml) of each flavonoids were used. In all cases, the antioxidant activity was dose-dependent. Rutin showed the highest inhibitory effect on RBC hemolysis than other flavonoid, that was 42.5. Also kaempferol, rutin, morin protect -SH groups 6%, 23.3%, 26.4% respectively. Results showed that flavonoids and plants contain flavonoids can be used as natural antioxidants for treatment and prevention of diseases which their pathogenesis are lipid peroxidation. | ||
