بررسی اثر جهش‌زایی غلظت‌های مختلف EMS و پرتو UV-C بر یونجه (Medicago sativa L.) | ||

| چکیده | ||
| بذر یونجه چندساله (L. Medicago sativa) توده همدانی پس از ضدعفونی، به‌مدت 24 ساعت در معرض غلظت‌های مختلف (EMS) Ethyl Methane Sulfonate قرار گرفت. پس از تیمار بذرها با EMS، در شرایط استریل بر روی محیط جامد ½ MS قرار داده شدند. پس از جوانه‌زنی طول ساقه‌چه، طول ریشه‌چه و تغییرات ظاهری نمونه‌های تیمار شده با غلظت‌های مختلف EMS مورد بررسی قرار گرفت. غلظت‌های کم EMS تأثیری در جوانه‌زنی بذرها نداشت، اما افزایش غلظت ماده یاد شده باعث کاهش تدریجی جوانه‌زنی گردید و در غلظت‌های بیشتر از 65 میلی‌مولارEMS، جوانه‌زنی متوقف شد. اثر منفی EMSبر رشد ریشه‌چه بیشتر از ساقه‌چه بود. بذرهای تیمار شده با 25 میلی‌مولار EMS، در شرایط هیدروپونیک کشت و تغییرات فنوتیپی در گیاهان M1 مورد ارزیابی قرار گرفت. از بین بیش از یک هزار گیاه M1، فقط در یکی از بوته‌ها تغییرات فنوتیپی مشاهده گردید. در گیاهان تیمار شده، تغییراتی از قبیل تغییرات در رنگ حاشیه برگها که حکایت از کاهش توان جذب فسفات داشت، مشاهده گردید. در بررسی گل آذین نمونه‌ها نارساییهای ظاهری از قبیل نقص در اندامهای جنسی مشاهده نگردید. در پرتوتابی بذرهای جوانه‌زده با نور UV-C، با طول موج 254 نانومتر با مقادیر متفاوت، تغییرات قابل مشاهده‌ای در گیاهان پرتوتابی شده، مشاهده نگردید. به منظور ایجاد جهش در یونجه، تیمار بذر با غلظت‌های مناسب EMS بهترین نتیجه را در تولید گیاهان موتانت ایجاد می‌نماید. در حالی‌که پرتو UV تأثیر قابل مشاهده‌ای در ایجاد جهش در گیاهان تیمار شده ندارد. | ||

| Seeds of alfalfa (Medicago sativa L.) were treated with various concentrations of Ethyl Methane Sulfonate (EMS) for 24 hours. The treated seeds were transferred onto 1/2 MS solid medium under aseptic condition. Germination percentage, shoot and root lengths and qualitative characteristics of the treated seedlings were evaluated. Low concentrations of EMS did not have adverse effects on seed germination. However, increase in concentration reduced the seed germination. At concentrations higher than 65mM EMS, germination as well as root and shoot growth were completely blocked. The inhibitory effect of EMS on root growth significantly was higher than on shoot growth. Seeds treated with 25mM EMS was grown under hydroponics condition and phenotypic changes of plants were evaluated. Only one plant exhibited phenotypic changes among treated population. Although leaf color change observed in one of the treated plants, other abnormalities were not observed in the population. The effect of UV-C on germinated seeds was evaluated via irradiation of the germinated seeds with 254nm wave length. At various doses of the UV light, however, phenotypic changes or inhibitory effects did not observe in the treated plants. In order to mutation induction in alfalfa, application of suitable concentrations of EMS gave the best results. However, irradiation of the germinated seeds with various doses of 254nm UV beam, did not affect the germinated seeds of alfalfa. | ||
