چکیده
Infectious bursal disease virus (IBDV) is the causative agent of Gumboro disease, an infectious disease of global economic importance in poultry. The expression of heterologous proteins in P.pastoris is fast, simple and inexpensive. In this study, VP2 encoding gene of classical D78 IBDV was amplified using reverse transcription (RT) polymerase chain reaction (PCR) and cloned into pPICZαA vector. Recombinant plasmid DNA was integrated into the chromosome of the transformed Pichia pastoris by electroporation and expressed protein identified by SDS- PAGE and western blotting. A recombinant protein was secreted into the supernatant from the yeast when induced with methanol. The expressed target protein in supernatant was bound with chicken anti IBDV Polyclonal antibodies. Western blotting with antibodies against D78 IBDV indicated that the recombinant VP2 protein retained its antigenicity. The concentration of secreted VP2 protein was 0.67mg/l.The production of recombinant VP2 protein indicated that P. pastoris was an efficient secreted expression system for D78 IBDV.

چکیده [English]
بیماری بورس عفونی، یکی از مهمترین بیماریهای ویروسی جوجه های صنعتی است. برای پیشگیری از بیماری، استفاده از واکسنهای معمول مرسوم است که با توجه به برخی محدودیتهای این واکسنها، در سال های اخیر بکارگیری واکسنهای نوترکیب در پیشگیری از این بیماری مورد توجه قرار گرفته است. در این خصوص بیان ژنهای پروتئین های ایمنی زا در سیستم مخمر پیشیا پاستوریس به شکل ویژه مورد نظر می باشد. در این مطالعه ژن VP2 از ویروس کلاسیک واکسینال D78 بوسیله روش RT-PCR تکثیر گردید و سپس در ناقل ترشحی pPICZαA کلون گردید و پس از آن مخمر پیشیا پاستوریس(سویه KM71H)توسط روش الکترو پوریشن بوسیله کلون مزبور ترانسفورمه شد. سپس کلونی های مخمر نوترکیب در یک « محیط غذایی مینیمال» تکثیر یافتند و بدین شکل پروتئین نوترکیب VP2 به جهت ترشحی بودن ناقل پلاسمیدی در مایه رویی بوسیله روش SDS-PAGE و Western blot شناسایی گردید. میزان بیان این پروتئین نو ترکیب 67/. میلی گرم در لیتر گزارش گردید. بیان پروتئین های ایمنی زا مانند VP2 در مخمر پیشیا پاستوریس به عنوان یک روش ساده، سریع و ارزان توصیه می گردد و با انتخاب کلون های مخمری مناسب تر و استاندارد نمودن پارامترهای مختلف کشت مخمری، میتوان میزان بیان این پروتئین ترشحی نوترکیب را افزایش داد.
