| چکیده | ||
| ترپسین از جمله آنزیم هایی است که در فرآیندهای بیوتکنولوژی کاربرد فراوان دارد. در آزمایشگاه کشت سلولی، طی فرآیند پاشاژ سلول ها، تریپسین جهت جدا کردن سلول های چسبیده شده به دیواره ظرف کشت سلولی استفاده می شود. همچنین جهت جدا کردن سلول ها از یک بافت می توان از تریپسین استفاده کرد. تریپسین در فرایند تولید پپتن و پروتئین هیدرولیز شده از کازئین شیر و سویا بطور انبوه و گسترده مورد استفاده قرار می شگیرد(21،1). تریپسین یک سرین پروتئاز است که در سیستم گوارش اکثر مهره داران یافت می شود ترپسین از جمله آنزیم هایی است که در فرآیندهای بیوتکنولوژی کاربرد فراوان دارد. در این مطالعه با استفاده از روش های رسوب دهی پروتئین ها با سولفات آمونیوم اشباع شده 35 درصد و 75 درصد، رسوب دهی با تری کلرو استیک اسید 5/2 درصد وزنی حجمی، رسوب دهی پروتئین ها در 5/6=pH و در نهایت با کروماتوگرافی تمایلی تریپسین موجود در پانکراس تخلیص شد. با این روش، از هر کیلو گرم پانکراس گوساله 8/2 گرم تریپسین خالص با فعالیت ویژه (U/mg) 155 استحصال گردید. نتایج این مطالعه نشان میدهد که با این روش می توان 26 درصد از آنزیم موجود در پانکراس را بدست آورد. در نهایت می توان گفت این روش یک روش ساده، سریع و مقرون به صرفه جهت تولید انبوه تریپسین باشد. | ||

| Trypsin is a serine protease found in the digestive system of many vertebrates, where it hydrolyses proteins. Trypsin has been used widely in various biotechnological processes. In this study trypsin was purified using two precipitation cut, 35% and 75% as saturation percent of Ammonium sulfate solutions, TCA precipitation (2.5% W/V), pH precipitation and finally affinity chromatography. This method could be used to obtain 2.8 g purified trypsin with specific activity 155 U/mg and 26% recovery from 1kg bovine pancreas. Therefore, this method is a simple, fast and commercial method for scale-up of trypsin extraction from bovine pancreas. | ||
