| در تحقیق حاضر آلودگی قارچی غذای مصرفی قزل آلای رنگین کمان با تاکید بر قارچ های بالقوه مولد مایکوتوکسین مورد ارزیابی قرار گرفت. نمونه ها از یک کارخانه تهیه خوراک دام و نیز از 6 مزرعه پرورش تهیه شدند. در کارخانه تولید خوراک از مواد اولیه تشکیل دهنده پلت شامل گندم، آرد گندم، سویا، پودر ماهی، گلوتن و نشاسته و نیز محصول نهایی و از مزارع پرورش قزل آلا از خوراک مصرفی نمونه گیری بر اساس دستورالعمل شماره 7570 موسسه استاندارد و تحقیقات صنعتی ایران صورت گرفت. نمونه ها در کیسه های پلاستیکی سربسته به آزمایشگاه منتقل شدند و بلافاصله رقت های مختلف تهیه شده از نمونه ها به منظور جداسازی قارچ به محیط های flavus Aspergillus و A.parasiticus AFPA)) دیکلران رزبنگال کلرامفنیکل آگار (DRCA ) منتقل گردید. کلنی های قارچی به محض ظهور بر روی محیط های کشت از طریق کشت مجدد بر روی محیط سابورو دکستروز آگار، خالص شده و با استفاده از تلفیقی از ویژگی های مورفولوژیک ماکرو و میکروسکوپی در سطح جنس و گونه مورد شناسایی قرار گرفتند. بر اساس نتایج بدست آمده، در مجموع 133 گونه قارچ از 11 جنس با فراوانی های مختلف جدا شد. بیشترین میزان آلودگی مربوط به محصول نهایی کارخانه (04/18درصد) و سپس به ترتیب آرد گندم (29/17درصد) و گلوتن (78/15درصد) بود. گونه های آسپرژیلوس از همه محل های نمونه گیری غیر از یک مورد گزارش شدند. جنس آسپرژیلوس با 88/54 درصد بالاترین فراوانی را دارا بود و بعد از آن به ترتیب جنس های سودالشریا (03/12 درصد)، پنیسیلیوم (52/10درصد) و آبسیدیا (77/9) قرار داشتند. در بین گونه های جدا شده بیشترین جداسازی مربوط به گونه.flavus A (84/36درصد) بود. از محصول نهایی کارخانه خوراک دام و مواد اولیه آن در مجموع 109 گونه قارچ از 11 جنس جدا شد. بیشترین میزان آلودگی مربوط به پلت (01/22درصد) و سپس آرد گندم (1/21درصد) و گلوتن (26/19درصد) بود. گونه های جنس آسپرژیلوس از همه نمونه های غذایی گزارش شدند، در حالی که جنس های سودالشریا و الوکلادیوم فقط از پلت جدا شدند. جنس آسپرژیلوس با 96/55 درصد بالاترین فراوانی را دارا بود و بعد از آن به ترتیب جنس های پنیسلیوم (84/12درصد)، آبسیدیا (01/11درصد) و سودالشریا (1/10درصد) قرار داشتند. سهم .flavus A 94/33درصد بود. در نمونه های به دست آمده از خوراک قزل آلا در 6 مزرعه پرورش در مجموع 24 گونه قارچ از 6 جنس جدا شد، که آسپرژیلوس به تنهایی 50 درصد از این آلودگی را تشکیل می داد که همگی مربوط به گونه .flavus A بود. سه مزرعه از غذای دست ساز و سه مزرعه از غذای آماده استفاده می کردند. میزان آلودگی در غذای دست ساز بیشتر از غذای آماده بود. | ||

| In the present study, fungal contaminations of trout feed were examined with special attention of potentially mycotoxigenic fungi. The feed samples were obtained from a factory and 6 farms. Sampling was done on final pellets and their ingredients including wheat, wheat flour, soya, fish powder, gluten and starch in the factory and in the farm according to the instruction of Institute of Standard and Industrial Research of Iran (No: 7570). Samples were transferred to the laboratory in nylon bags and cultured for fungal isolation on specific media i.e. Aspergillus flavus and parasiticus agar (AFPA) and Dichloran rosebengal chloramphenicol agar (DRCA). Fungal colonies were purified from primary cultures by sub-culturing on Sabouraud dextrose agar plates and identified at the genus or species level by a combination of macro- and microscopic morphological criteria. Based on the results obtained, a total of 133 fungal isolates belonging to 11 different genera were isolated. The most contamination occurred on the factory’s final production stage, followed by the wheat flour (17.29%) and gluten (15.78%). Aspergillus species were isolated from all the samples except one. The genus aspergillus was the most prominent fungus isolated (54.88%) followed by the genera penicillium (10.52%) and Absidia (9.77%). Aspergillus flavus was most frequently isolated fungus (36.84%). A total number of 109 fungal species from 11 genera were isolated from the pellets and feed ingredients in the factory. The most contamination was in the pellet (22.01%), followed by wheat flour (21.1%) and gluten (19.26%). Aspergillus species were present in all the samples, while pseudallscheria and ulocladium were isolated only from the pellets. Aspergillus was the most prominent genus (55.99%), followed by the genera penicillium (12.84%), absidia (11.01%) and pseudallscheria (10.1%). From the farm samples, a total of 24 fungal species were isolated, of which 50% were belonging to the genus aspergillus from the species, Aspergillus flavus. Three of the farms used handmade feed while the other three farms consisted of factory made feed. The feeds that were handmade were more contaminated than that of the factory made feed. | ||
