| بیماری زبان آبی یا بلوتانگ از جمله بیماریهای عفونی است که عمدتا توسط حشرات منتقل و باعث بروز ضایعاتی در مخاط بینی،دهان و اندام حرکتی و نیز سقط جنین در نشخوارکنندگان می شود.هدف از این مطالب جستجوی ژنومی-ویروس بلوتانگ در جنینهای سقط شده گوسفند به عنوان عامل سقط بود.به منظور جستجوی ژنوم ویروس بلوتامگ، از تعداد 50 بره ی سقط شده نمونه ی خون ،کبد و طحال تهیه شد.به نمونه های خون EDTA اضافه شد و با استفاده از ریمازول استخراج RNAانجام پذیرفت.نمونه ها به روش RT_PCR مورد آزمون قرار گرفتند.نتایج حاصل از آزمون در ژل آگاروز 1.5 درصد،الکتروفورز و با اتیدیوم بروماید رنگ آمیزی گردید و توسط اشعه UV قرائت شد.در هیچ یک از نمونه ها نتیجه مثبت مشاهده نشد، به جز یک مورد که برای تعیین توالی ارسال گردید ولی نتیجه تعیین توالی،ویروس بلوتانگ را تایید نکرد. بنابراین مطالعه ی حاضر میتواند گویای آن باشد که در موارد بررسی شده در مطالعه حاضر،احتمالا عفونت با ویروس بلوتانگ عامل سقط نبوده است اما ممکن است در صورت بالا بردن حجم نمونه و اجرای نمونه گیری به طور پراکنده در سرتاسر سال بتوان حتی در مناطق مورد مطالعه در طرح حاضر حضور ویروس یا ژنوم آن را نشان داد. | ||

| Detection of bluetongue virus in aborted lamb fetuses in Chaharmahal va Bakhtiari and Isfahan provinces, by RT-PCR method Background: Bluetongue or muzzle sore disease is an infectious disease which is mainly transmitted by insects. It can cause erosions in mouth, nasal mucosa and limbs and also abortion in ruminants. The aim of this study was genomic detection of bluetongue virus in sheep aborted fetuses as an aborting agent. Materials and methods: Samples were collected from blood, liver and spleen of 50 aborted lambs. EDTA was added to blood samples. RNA was extracted by using Rimazol. cDNA was synthesize was detected by RT-PCR. All products were separated by 1/5% agarose gel electrophoresis and stained in ethidium bromide. The results of all samples were negative, except one of them which was sent for sequencing and the result of sequencing didn't confirm presence of this virus. This study shows that Blue tongue virus does not cause abortion in ewes in studied areas of Iran, but it is possible that by increasing the sample size and sampling during the year in these areas, the presence of virus or its genome can be detected too. | ||
